What is the FDA Orange Book?

The Orange Book identifies drug products approved by FDA on the basis of safety and effectiveness under the Federal Food, Drug, and Cosmetic Act and provides related patent and exclusivity information. FDA describes therapeutic equivalence evaluations as public information and advice for state health agencies, prescribers, pharmacists, and others involved in drug product selection.

The publication is specific to the U.S. FDA regulatory framework. It is not a global register of medicines or a substitute for another country’s approval, substitution, patent, or exclusivity system.

The official Orange Book includes product information organized around applications and products. The FDA’s searchable Electronic Orange Book can be queried by active ingredient, proprietary name, applicant, application number, dosage form, route of administration, or patent number. FDA also publishes downloadable data files and provides Orange Book data through the openFDA API in machine-readable JSON.

The downloadable files contain product, patent, and exclusivity information. FDA describes them as ASCII text files separated by a tilde (~), with field definitions published alongside them. Application and product numbers help researchers connect product records with patent or exclusivity records.

FDA states that downloadable Orange Book data files are updated monthly. The Electronic Orange Book page also displays a current-through date. As of August 2026, the page states that its information is current through August 2026. Because listings and status information can change, check the FDA page and current file date before relying on a record for time-sensitive work.

What information does Orange Book drug data contain?

At the product level, Orange Book drug data describes characteristics such as:

  • Ingredient: The active ingredient or ingredients. FDA’s field description states that multiple ingredients are listed alphabetically and separated by a semicolon.
  • Dosage form: The product form.
  • Route of administration: The route associated with the product.
  • Strength: The strength for the product.
  • Application number: The FDA-assigned number for the application.
  • Product number: The FDA-assigned number identifying a product within the application. FDA states that each strength is a separate product and that product numbers may repeat for multiple-part products.
  • Approval date: The date associated with approval in the product information.
  • Therapeutic equivalence code: A code with a defined FDA regulatory meaning.
  • RLD: Whether the product is identified as a reference listed drug.
  • Reference standard: A designation used in generic drug development and bioequivalence reference products.
  • Patent: Patent records associated with an application and product.
  • Exclusivity: FDA-granted exclusivity information associated with a product and application.

These fields answer different questions. Ingredient, dosage form, route, and strength describe the product. Application and product numbers provide regulatory identifiers. Therapeutic equivalence, RLD, and reference standard fields describe regulatory relationships. Patent and exclusivity fields describe information recorded in the relevant Orange Book sections.

Therapeutic equivalence: what the codes mean

Therapeutic equivalence is an FDA evaluation applied to certain multisource prescription drug products approved under the Federal Food, Drug, and Cosmetic Act. FDA’s guidance explains the agency’s approach to therapeutic equivalence evaluations and therapeutic equivalence codes, commonly called TE codes.

A TE code communicates a defined regulatory evaluation under FDA’s framework. Researchers should consult FDA definitions and guidance for the precise meaning of a particular code rather than treating all codes as interchangeable. Therapeutic equivalence evaluations do not, by themselves, guarantee identical patient outcomes.

The Orange Book distinguishes products by dosage form and route. FDA’s FAQ explains that an ingredient search presents products by dosage form and route grouping. Within such a grouping, an RLD is identified for the innovator product. Other approved generic products may have a TE code. The relevant code should therefore be read with the product’s ingredient, dosage form, route, strength, application, and reference designations.

FDA also states that over-the-counter products in the OTC section are not evaluated for therapeutic equivalence. An OTC listing should not be interpreted through the same TE-code framework used for evaluated products in the applicable prescription sections.

Two fields that are often confused are reference listed drug (RLD) and reference standard.

An RLD is the drug product identified in the Orange Book as the reference product for therapeutic-equivalence purposes. FDA’s FAQ describes the RLD as the innovator product within a dosage-form-and-route grouping.

A reference standard is the product an abbreviated new drug application (ANDA) applicant uses to establish bioequivalence. The Orange Book Preface gives an example involving levothyroxine: a product may be selected as the reference standard for ANDA applicants even where the relationships among RLDs and therapeutic-equivalence ratings require closer examination.

These designations can overlap, but they are not synonyms. A product’s RLD status and reference-standard status should be read from their respective fields and the associated FDA explanations. Do not replace one designation with the other when building a research dataset or interpreting a product relationship.

How drug patents and exclusivity fit into the listing

The Orange Book also contains information about drug patents and exclusivity. These records are separate from therapeutic equivalence evaluations, although they may be connected through application and product identifiers.

FDA’s data descriptions state that patent records can include:

  • Patent number
  • Patent expiration date as submitted by the application holder, including applicable extensions
  • Drug substance and drug product flags for certain patents submitted after August 18, 2003
  • Patent use code
  • Patent delist request flag
  • Patent submission date

The data files also describe exclusivity records using an application type, application number, product number, exclusivity code, and exclusivity date. The exclusivity code designates exclusivity granted by FDA to a drug product, while the exclusivity date identifies when that exclusivity expires.

A patent entry is a recorded patent listing with associated fields; it is not, by itself, an interpretation of enforceability, infringement risk, or the outcome of a dispute. Researchers examining commercial or regulatory strategy should read FDA definitions, relevant statutes, and current legal materials separately.

Worked example: reading one Orange Book product record

Consider the FDA’s documented example involving Mylan’s levothyroxine product, ANDA 076187, and Thyro-Tabs.

Start with the application number. 076187 identifies the ANDA associated with Mylan’s levothyroxine product in the example. That number helps connect the product to application-level information; it is not a universal identifier for every levothyroxine product.

Next, read the product context. The record concerns levothyroxine, but the ingredient alone is not enough to compare products. A researcher should also examine dosage form, route, strength, product number, and the relevant reference designations. FDA explains that each strength is a separate product, so a product-level analysis should not collapse all strengths into one record.

FDA states that Mylan’s product was selected as the reference standard for ANDA applicants to use to establish bioequivalence to Thyro-Tabs. This describes the role FDA assigned to that product in the ANDA bioequivalence process.

The example also describes the therapeutic-equivalence relationship. If an ANDA using Mylan’s levothyroxine product as its reference standard is approved, the ANDA can receive an AB4 rating. The example further states that an ANDA applicant may obtain an AB rating to other RLDs—Unithroid, Synthroid, and Levoxyl—by submitting supplements demonstrating bioequivalence to those other RLDs and satisfying the other therapeutic-equivalence criteria.

What does this record tell a researcher?

  1. It identifies an ANDA and a product associated with levothyroxine.
  2. It shows that FDA assigned the product a reference-standard role for a particular bioequivalence pathway.
  3. It explains how an AB4 rating may relate to that reference-standard choice if the relevant ANDA is approved.
  4. It shows why RLD and reference standard must be analyzed as separate fields.

It does not establish that every levothyroxine product has the same dosage form, route, strength, RLD relationship, reference standard, or TE code. It also does not answer whether a particular package is currently marketed or available in a specific country. Those questions require appropriate current official information and, where relevant, additional sources.

Key limitations to keep in mind

The Orange Book is an official FDA publication within the U.S. drug-approval framework, but it is not a universal source for every type of drug information. FDA’s FAQ states that some information users may seek is not found in the Orange Book and directs them to other FDA channels for more specific information.

The dataset’s fields should be interpreted at their stated level. An application number identifies an FDA application, and a product number identifies a product within that application. Neither identifier alone establishes a manufacturer’s current commercial activity, a product’s availability in a particular market, or a clinical outcome.

Patent and exclusivity entries require the same care. Their presence supplies recorded regulatory information, while questions about legal effect or current status may require additional official and legal sources. Review the current FDA files, definitions, and applicable materials before using the data for a decision that depends on changing information.
